GPT/ALAT catalyzes the transformation of 2-Oxiglutarate and L-Alanine into L-Glutamate and Pyruvate. Subsequently, the Lactate Dehydrogenase (LDH) catalyzes the transformation of the pyruvate formed and of the NADH into L-Lactate and NAD+ producing a decrease in Absorbance (Abs) of the reaction which is measured fotometrically at a wavelength of 340 nm. The rate of decrease of the Abs is proportional to the activity of the enzyme GPT/ALAT in the sample.

Alanine + 2-Oxoglutarate     ( ALAT (GPT)  → L-Glutamate + Pyruvate

Pyruvate + NADH + H+        ( LDH)    →    D-Lactate + NAD+
